After sequencing the human genome a decade ago, researchers have continued their projects, but now to try to better understand how, and when, genes are expressed in health and disease. Efforts have been concentrated on the measurement of the expression of RNA transcripts. In an analogy to the genome, the term "transcriptome" was created to refer to the complete set of RNAs in a cell type or tissue in a particular situation. Transcriptomics is the science that studies this issue and it is a branch of functional genomics. Transcriptomics in Heath and Disease provides a comprehensive overview of the science of transcriptomics initially in health, focusing on the concept of the transcriptome and...

Oncolytic Viruses - Genetically Engineering the Future of Cancer Therapy

The ability to genetically engineer oncolytic viruses in order to minimize side effects and improve the selective targeting of tumor cells has opened up novel opportunities for treating cancer. Understanding the mechanisms involved and the complex interaction between the viruses and the immune system will undoubtedly help guide the development of new strategies. Theranostic biomarkers to monitor these therapies in clinical trials serve an important need in this innovative field and demand further research.

Tailoring NK Cell Receptor-Ligand Interactions: an Art in Evolution, 2nd Edition

Recognition and killing of aberrant, infected or tumor targets by Natural Killer (NK) cells is mediated by positive signals transduced by activating receptors upon engagement of ligands on target surface. These stimulatory pathways are counterbalanced by inhibitory receptors that raise NK cell activation threshold through negative antagonist signals. While regulatory effects are necessary for physiologic control of autoimmune aggression, they may restrain the ability of NK cells to activate against disease. Overcoming this barrier to immune surveillance, multiple approaches to enhance NK-mediated responses are being investigated since two decades. Propelled by considerable advances in the un...

Thymus Transcriptome and Cell Biology

This volume focuses on a challenging field in biomedicine: the genetic control of central immune tolerance. The thymus gland is a lymphoid organ implicated in T cells' maturation, differentiation, and selection. Its function is associated with the control of immune homeostasis in the body, establishing central immune tolerance, and preventing the onset of autoimmune diseases. This book focuses on thymus development, their cellular components and their respective function, and the peculiar gene expression profiling (transcriptome) found in the medullary thymic epithelial cells (mTECs) that are implicated in the self-representation in the thymus and the Autoimmune regulator (Aire) gene. Chapters also explore the mutations in the Aire gene, manifestation of autoimmune diseases, and the role of cell-cell interactions within the thymus with implications in the negative selection (elimination) of nascent autoreactive T cells in preventing aggressive autoimmunity. This new edition includes two new chapters devoted to the genome editing of the Aire gene through Crispr-Cas9 system, and thymic involution. All chapters have been updated to reflect the latest research in the field.
