Alzheimer's dementia (AD) affects 6 million Europeans with 10% of people over age 65 and more than a quarter over 85. Given the steady aging of European societies, dementia and cognitive decline have developed into a major health problem with an enormous socioeconomic impact for patients, their families and caregivers, national health care systems, and society. Without any means to prevent or delay disease onset, the number of people with dementia is predicted to double by 2030 and triple by 2050. There is an urgent need for innovative strategies to increase understanding of pathological events that would translate into the development of successful prevention or, possibly, novel treatment strategies. Progresses in understanding pathological events in AD have been possible by using cell cultures, genetically modified organisms and animal models that lack the complexity of events occurring in humans. We need to overcome this limitation also by using data from humans - for studying pathological pathways in AD in a multidisciplinary setting.
Alzheimer’s disease (AD) is a chronic neurodegenerative disorder characterized by progressive cognitive dysfunction and memory loss, inability to perform the activities of daily living and mood disorders. According to the so-called “amyloid cascade hypothesis”, amyloid-ß- peptide (Aß), produced by beta- and gamma- secretase-mediated cleavages of the amyloid precursor protein (APP), plays a pivotal role in the pathogenesis of AD. Aß was also shown to contribute to AD pathology by stimulating the hyperphosphorylation of tau which is responsible for the formation of neurofibrillary tangles. Brain function is under metabolic control, which in turn determines the equilibrium of homeostatic systems that affect neuronal and glial networks on the molecular, cellular, and systems levels. The collection of articles ranges from molecules and mechanisms involved in regulating homeostasis and neuronal excitability to therapeutic mechanisms tailored to restore homeostatic function. It also features neurological diseases and novel treatment approaches that are based on metabolic and homeostatic interventions. Together, the collection of articles outlines novel strategies to restore brain function in neurology and highlights limitations of conventional pharmacological approaches. We suggest that restoration of molecular and biochemical networks could lead to a new era of therapeutic opportunities.
Frontiers in Pharmacology was launched in 2010, with a number of sections which were eventually reorganized. The founding Field Chief Editor was Prof. Théophile Godfraind, an eminent scientist active in cardiovascular pharmacology, who pioneered the discovery of calcium antagonists. At that time he invited me to serve as Chief Editor for a section named “Analytical and Experimental Pharmacology”. Later on, our section enlarged and was re-named as “Experimental Pharmacology and Drug Discovery” to outline the translational potential of fundamental pharmacological research and theoretical analysis to the improvement of human health, through the invention of novel medicinal products. We are now entering the 10th year of editorial activity, which sees the publication of the 1,000th paper in our section. Such an achievement is very rewarding for us and our community, but it is even more remarkable when placed into the timeline of our development. In fact, in a 10-year frame we have significantly grown in quantity and quality, e.g. both in number of published papers and in scientific impact. Intimate and often unseen, the sketchbook means something different to each illustrator. It might be a beautiful object, a work of art in its own right, where every line is painstakingly considered. It might be a pictorial playground, where mistakes can make art. The boundaries between sketchbooks, notebooks and visual journals are often blurred, lending to the creativity that fills their pages. It is likely that you will recognize many of the illustrators featured, including classic childhood favourites Beatrix Potter, Jean de Brunhoff, Edward Ardizzone and Tove Jansson, and established names such as Beatrice Alemagna, Oliver Jeffers and Shaun Tan. Others are up-and-coming, for example Charlotte Ager and Leah Yang. Martin Salisbury draws on decades of experience as an illustrator and educator to shed light on the lives and work of each artist. He even reveals pages from his own sketchbooks, exposing the rawness of his ideas and the narratives that surround them.
