The first half of this thesis describes the development of a ligation based assay which can be used for systematic detection of 2'-O-methyl modifications in ribosomal RNA and its subsequent application in studies related to effect of ribosome inhibiting drugs on rRNA modification in yeast. This detection method is based on the enzymatic ligation of oligonucleotides using the modified or unmodified RNA as the template. Multiple modification sites can be examined in the same ligation reaction making this a great tool for high throughput detection of RNA modifications.
